The Organoid and Tissue Modeling Shared Resource (OTMSR) provides cutting-edge services and expertise in in vitro 3D tissue culture modeling to researchers across the CU Anschutz campus. In this role, you will work with human 3D organoids, CRISPR/Cas9 gene editing tools, and engineered stem cell lines. Daily responsibilities will include the generation and culture of organoid cell lines, the execution and optimization of experimental protocols, and the use of advanced imaging and molecular tools.

Key Responsibilities:

  • Generation, maintenance, characterization, and biobanking of organoids produced from mouse and human-derived tissues.

  • Culture and differentiation of human pluripotent stem cells.

  • Genetic manipulation of pluripotent stem cells, including CRISPR/Cas9 gene editing (point mutation, knock-in, knock-out).

  • Molecular biology (PCR, electrophoresis, plasmid restriction/ligation, bacterial transformation/culture/purification).

  • Execution of cell-based assays - clonogenic assays, multi-color fluorescence, 3D, live cell, and confocal imaging.

  • Gene expression analysis (qPCR, RNAseq, scRNAseq, flow cytometry analysis, and sorting).

  • Maintain mouse colonies (husbandry, genotyping, tissue processing, etc.).

  • Development of new techniques/assays associated with organoid and iPSC culture systems.

  • Will occasionally assist in the Molecular Core of the Diabetes Research Center (Sanger sequencing, mycoplasma testing, DNA purification, cell line authentication, etc.).

  • Able to work independently and closely with a team.

  • Perform general laboratory maintenance, including ordering supplies, receiving laboratory reagents, making stock solutions, cleaning/maintaining laboratory space and equipment, etc.

Work Location:

Onsite - this role is expected to work onsite and is located in Aurora, CO.

Why Join Us:

The Organoid and Tissue Modeling Shared Resource (OTMSR) at CU Anschutz provides a stimulating environment for mastering 3D cell culture, genetic engineering, and stem cell differentiation protocols. In this role, you will utilize cutting-edge cell culture techniques, gene editing tools, and advanced instrumentation to develop novel in vitro models to study complex human development and disease. Skills and experience attained in this role are well-suited for those considering pursuing post-graduate school.
